Design Notes

1. Both caga and SpyTag are gene sequences that I have chosen because they have been successfully expressed in E. coli, ensuring their successful expression in my experiment. 2. The choice of GST purification tag is due to the fact that another module in my project uses an His tag. For the subsequent CoIP (co-immunoprecipitation) experiment to verify the interaction between the two, I need to use two different tags to confirm their binding.

Source

csgA is a gene in the E. coli genome, while spytag is a gene on the Streptococcus pyogenes genome.
